New Publication: Unlocking Interneuron Development with 3D Culture Systems

We are excited to share our latest paper in Science Advances! In this study, we demonstrate direct lineage reprogramming of human glial cells into GABAergic interneurons using a three-dimensional (3D) culture system.


Within just two weeks, single-nucleus RNA sequencing revealed distinct neuronal clusters, including a parvalbumin (PV) subtype with features of chandelier interneurons—a key player in cortical circuitry. Our trajectory analysis uncovered the stepwise conversion from glia to neuron and identified previously unknown genes critical for PV fate and synaptic development.


This breakthrough highlights the potential of human brain cell engineering for repairing circuits in neurological disorders and opens new avenues for regenerative strategies.
